Shodex ligand exchange chromatography tutorial

by | Glycans, Liquid Chromatography

Shodex_ligand_exchangeThis video describes the separation mode of ligand exchange (LEX). The main application is the complex analysis of sugars. The LEX columns always have a double separation mode. This means two different separation modes in one column. Ligand exchange and size exclusion is working together in order to get the best separation.  The separation is based on differences in the analyte’s coordination complex. The stationary phase is modified with metal sulfonate complex ions. Examples for often used cations are Ca2+, Na+, Zn2+,Pb2+. With this separation mode combination, even sugars with the same molar mass can be separated because different stable complexes can be built.

The video shows the detailed theory as well as the products and why and how they can used for sugar analysis. The columns are polymer based for maximum stability.
